چکیده

RBCs from patients with hemolytic anemia due to pyruvate kinase (PK) deficiency are characterized by a decreased total adenine and pyridine nucleotide content. Because phosphoribosylpyrophosphate (PRPP) is a precursor of both adenine and pyridine nucleotides, we investigated the ability of intact PK-deficient RBCs to accumulate PRPP. The rate of PRPP formation in normal RBCs (n = 1 1 ) was 2.89 ± 0.80 nmol/min . ml RBCs. In contrast, the rate of PRPP formation in PK-deflcient RBCs (n 4) was markedly impaired at 1 .03 ± 0.39 nmol/min . mL RBCs. Impaired PRPP formation in these cells was not due to the higher proportion of reticulocytes. To study the mechanism of impaired PRPP formation, PK deficiency was simulated by

Selective expression of phosphoribosylpyrophosphate synthetase superactivity in human lymphoblast lines.

Phenotypic expression of 5-phosphoribosyl 1-pyrophosphate (PRPP) synthetase superactivity was examined in lymphoblast lines derived from six unrelated male patients. Fibroblasts from these individuals have increased rates of PRPP and purine nucleotide synthesis and express four classes of kinetic derangement underlying enzyme superactivity: increased maximal reaction velocity (catalytic defect)...

Decrease in subunit aggregation of phosphoribosylpyrophosphate synthetase: a mechanism for decreased nucleotide concentrations in pyruvate kinase-deficient human erythrocytes.

Pyruvate kinase (PK)-deficient RBCs have several unexplained metabolic abnormalities, such as decreased concentrations of total adenine nucleotides (AMP, ADP, and ATP) and total (oxidized and reduced) nicotinamide adenine dinucleotide (NAD). Because 5-phosphoribosyl-1-pyrophosphate (PRPP) is an intermediate in the synthesis of adenine nucleotides and NAD, we investigated PRPP synthetase (PRPPS)...

ENZYME LEVELS IN THE SERA AND ERYTHROCYTES OF PATIENTS WITH VISCERAL LEISH MANIASIS

Twenty five blood samples from 6-month to 5-year old children with visceral leishmaniasis were analysed for various enzymes. Aspartate transaminase (AST), alanine transaminase (ALT), lactate dehydrogenase (LD) and its various isoenzymes were estimated in the serum, while glucose 6-phosphate dehydrogenase (G6PD) was measured in red blood cells. For comparison, blood samples from healthy chi...

Secondary Metabolite Contents and Antioxidant Enzyme Activities of Cichorium intybus Hairy Roots in Response to Zinc

Hairy root systems are formed by transforming plant tissues with the “natural genetic engineer” Agrobacterium rhizogenes. In most plants such as Cichorium intybus L., hairy root cultures have proven to be an efficient system for secondary metabolites production. The effect of Zinc (ZnSO4), a heavy metal, was investigated at different concentrations (0, 1, 5 and 10 mM) on some secondary metaboli...
